Obviously, Hisoka and the other Hunters we saw during the exam used Nen, but they weren't separate from the characters who couldn't use Nen, so they only gave the audience a small hint of what was going on and it makes it really interesting to go back and re-examine certain scenes. In addition, the way Heaven's Arena is used to introduce Nen, it shifts from the usual shonen trope of "Hisoka has a superpower so Gon has to learn a superpower to beat him" to "Gon and Killua have reached the physical limit of what they can accomplish as fighters and the next step in their training is to learn a superpower." Hisoka's gatekeeping of floor 200 makes it explicit that the issue isn't that they need to train to beat an external opponent, this is all part of their personal development.
